For the uprated R50 version, VW (or Aisin) apparently made some part enhancements to the transmission to handle the additional torque of the tuned motor. I forget the torque specs of the R50 (850 Nm seems to ring a bell, but I'm not sure), but if you're producing more than that on a stock V10 tranny, I'd say if you toe in to that newfound power too often you'll be replacing the tranny. I'd start setting aside money now too, as you've just voided the tranny warranty by adding more juice to the engine.
